Fybrix is a synthetic, animal-free and easy to use cell culture matrix. It is mechanically and biochemically easy to tailor.
Fybrix works similar to BME but more controlled and cell extraction is dead easy: cooling for a minute and centrifugation.
Fybrix was developed as the synthetic PIC gel at Radboud University (Nature 2013). It finds application as a synthetic and animal-free cell culture matrix.
Due to its structural similarity to biological materials, such as collagen and fibrin, Fybrix has unique biomimetic properties. As synthetic material, Fybrix gels have a controlled composition and prepared reproducibly, The biochemical properties are tuned by introducing a range of (bio)molecules, including peptides (like GRGDS), proteins, glycans, dyes, crosslinkers, etc.

Moreover, Fybrix gels have unique thermal properties: gelation is induced by heating to 37 °C; cooling back to 5 °C instantly removes the gel, massively simplifying cell extraction for down stream analysis.
Fybrix gels have been successfully used to culture stem cells, fibroblasts, epithelial cells, smooth muscle cells, endothelial cells, nerve cells, hepatocytes, kidney cells, immune cells, different cancer cells, and preosteoblasts. In addition, PIC gels have been applied to grow mammary gland organoids, liver organoids, prostate organoids and salivary gland organoids. Many other organoid studies are currently going on. To make 3D Fybrix cell cultures, simply mix the polymer solution with the cell suspension at 5 °C and heat place the microplate in the incubator. After the experiment, the gel is dissolved by cooling and cells can be isolated by centrifugation, ready for down stream analysis. No chemicals necessary! Fybrix is a unique material. It combines the reproducibility and customizability of synthetic materials with the properties of biological materials. Compared to other synthetic gels, Fybrix has excellent biological properties. Compared to biological materials, Fybrix benefits from a controlled composition and easy cell retrieval.
